Genomic profiling of Mixer and Sox17β targets during Xenopus endoderm development

Abstract: The transcription factors Mixer and Sox17␤ have well-characterized roles in endoderm specification during Xenopus embryogenesis. In order to more thoroughly understand the mechanisms by which these endodermal regulators act, we expressed Mixer and Sox17␤ in naïve ectodermal tissue and, using oligonucleotide-based microarrays, compared their genomic transcriptional profile to that of unaffected tissue. Using this approach, we identified 71 transcripts that are upregulated by Mixer or Sox17␤, 63 of which have pr… Show more

“…Among these genes, sp8 is transiently expressed in the OV in mouse embryos (15) so we analyzed expression patterns of sp8 in X. tropicalis by in situ hybridization in WT and mutant embryos. As also reported for EST10 (19), staining for sp8 mRNA was found in the olfactory bulb, midbrain, hindbrain, and the OV in WT embryos, whereas no such signals were detected in ecl mutants ( Fig. 1 E-H).…”

“…Limb/fin outgrowth in chick and zebrafish embryos also employs sp8 (18), suggesting the conserved function of sp8 across phylogeny. In Xenopus, the gene was identified as a target of sox17 (19). Here, we demonstrate a role for sp8 in inner ear development.…”

“…1H,O). There may also be a difference in regulation between endodermal transcripts, which reflects the intricate network of gene regulation involved in endoderm formation reported in array experiments (Dickinson et al, 2006;Sinner et al, 2006).…”

“…Over the past few years extensive investigation has contributed to the elucidation of the molecular mechanisms underlying initial endoderm formation (Dickinson et al, 2006;Ober et al, 2003;Sinner et al, 2006;Tam et al, 2003;Taverner et al, 2005). However, we still know relatively little about subsequent endodermal patterning and how different domains of the endoderm progressively become specialized to generate distinct organ primordia.…”
