Germ cell sex prior to meiosis in the rainbow trout

Abstract: Germ cells make two major decisions when they move from an indeterminate state to their final stage of gamete production. One decision is sexual commitment for sperm or egg production, and the other is to maintain mitotic division or entry into meiosis. It is unclear whether the two decisions are made as a single event or separate events, because there has been no evidence for the presence of germ cell sex prior to meiosis. Here we report direct evidence in the fish rainbow trout that gonia have distinct sexua… Show more

“…In the fish species examined in detail by in situ hybridization (ISH), vasa expression occurs in both mitotic and meiotic germ cells of both sexes 27 , 32 . A similar observation has been described also for other germ genes such as boule and dazl in medaka 32 and dazl in trout 9 . One exception is boule , whose expression takes place in male gonia but not in female gonia 9 .…”

“…This distinction is usually made on the basis of difference of DAPI staining between mitotic oogonia and meiotic oocytes. This dye does not stain the oocyte nucleus in many teleost species including medaka 32 and trout 9, 39. Another dye is propidium iodide, which produces a similarly differential staining in zebrafish 29, gibel carp 27.…”

“…Boule is the only DAZ family member in invertebrates including Drosophila 20 and C. elegans 21. In the rainbow trout, differential expression of boule and dazl reveals germ cell sex prior to meiosis2223.…”

“…The samples were finally stained for nuclei by using DAPI (µg/ml) and embedded in the Gold Antifade reagent (Invitrogen) for microscopy. In this FISH procedure, two differently labeled antisense RNA probes are co-incubated with a sample in the hybridization step and subjected to sequential color development into green and red fluorescence 62.…”
