GFP-Forked, a genetic reporter for studyingDrosophilaoocyte polarity

Abstract: The polarized organization of the Drosophila oocyte can be visualized by examining the asymmetric localization of mRNAs, which is supported by networks of polarized microtubules (MTs). In this study, we used the gene forked, the putative Drosophila homologue of espin, to develop a unique genetic reporter for asymmetric oocyte organization. We generated a null allele of the forked gene using the CRISPR-Cas9 system and found that forked is not required for determining the axes of the Drosophila embryo. However, … Show more

“…This could be due to the efficacy of the RNAi-transgene, or suggestive of a yet-to-be identified negative regulator acting on PGRP-LB. Given previous characterization of the Mata-Gal4, we do not believe the finding is a result of GAL4 leakiness, as GFP expression is restricted to the ovary (Baskar et al 2019). However, transgene expression elsewhere in the adult or embryo not examined may potentially yield a phenotypic effect.…”

“…Oogenesis and development is a complex process regulated by multiple genes. In Drosophila , an organism where oogenesis and development are well understood, oogenesis begins with the asymmetric division of the germline stem cells . The Nudel gene is maternally required both for structural integrity of the egg and for dorsoventral patterning of the embryo .…”
