Glabridin from Glycyrrhiza glabra Possesses a Therapeutic Role against Keloid via Attenuating PI3K/Akt and Transforming Growth Factor-β1/SMAD Signaling Pathways

Abstract: Glabridin (Gla) is a typical flavonoid isolated from the Glycyrrhiza glabra with various bioactivities and is a common additive in many cosmetics. In our study, we evaluated the antiscarring effect of Gla from G. glabra in a rabbit ear hyperplastic scar model. Hematoxylin and eosin staining and Masson staining were applied to determine the pathological changes and collagen fibers of scar tissue in rabbits. The results suggested that Gla could reduce rabbit ear scar hyperplasia, inhibit inflammation, and decrea… Show more

“…The hydrophilic fraction of liquorice extracts is known to have anti-inflammatory effects [85]. Glabridin's effectiveness against keloid fibroblasts was shown by Zhang et al [86]'s experiment. Glabridin is a flavonoid obtained from the plant Glycyrrhiza glabra that possesses a variety of biological characteristics.…”

“…Glabridin was also found to inhibit the production of collagen. The cause was shown to be the suppression of the PI3K/Akt and TGFβ-1/SMAD pathways [86]. Liquorice could be a potential therapeutic agent for keloid treatment with further research.…”

“…Several non-canonical TGF-b pathways are involved in keloid formation. These include the MAPK (94,198,199), ERK 1/2 (44,200) phosphatidylinositol-3-kinase (P-I3K)/AKT (44,104,(200)(201)(202), c-Jun amino-terminal kinase (94), p38 mitogen-activated protein kinase (p38/MAPK) (94,(203)(204)(205), and Rho-like (82) signaling pathways. The multi-kinase inhibitor sorafenib induces cell arrest of keloid fibroblasts by blockade of the intracellular TGF-b/Smad and MAPK/ ERK pathways (206).…”

“…Therefore, alternative approaches are urgently needed, natural products have always held a privileged position as valuable sources and inspirations for new drug development ( Newman and Cragg, 2020 ). Some Chinese medicines show promising anti-fibrotic effects ( El-Tantawy and Temraz, 2022 ; Zhang et al, 2022a ).…”
