GLB‐13 is associated with oxidative stress resistance in caenorhabditis elegans

Abstract: Globins constitute a superfamily of heme-binding proteins that is widely present in many species. There are 33 putative globins in the genome of Caenorhabditis elegans, where glb-13 is a homolog of neuroglobin (Ngb) based on sequence analysis and specific expression in neurons. Here we examined whether glb-13 as well as Ngb is also associated with resistance to reactive oxygen species (ROS) induced by paraquat. Our results showed that the mRNA level of glb-13 was significantly upregulated after paraquat exposu… Show more

“…neuronal cells from hypoxia/ischemia, ROS/NOS, or β -amyloid peptides ( Jin, et al., 2008 ; Liu, et al., 2009 ; Ren, et al., 2013 ). Consistently, our data show that Ngb is significantly increased at 7 and 14 days after combined remote ischemic conditioning, compared with ischemic control group suggesting that Ngb may be involved in PerC+PostC-mediated neuroprotection after focal ischemia.…”

“…elegans shows 33 globins, diverse in gene and protein structure being localized mostly in neurons (Tilleman et al, 2011). Interestingly, globin-5 and globin-13 have been reported to be homologous to Ngb (Abergel et al, 2016;Kodama-Namba et al, 2013;Ren et al, 2013). Globin-5 is involved in the cross-modulation of homeostatic responses to O2, CO2, and temperature; in particular, the interaction of globin-5 with soluble guanylate cyclase-35 is pivotal for the adaptation to either low or high O2 levels (Abergel et al, 2016;Kodama-Namba et al, 2013).…”

“…According to the methods of Xu 16 and Ren et al , 19 the lifespan assay was carried out after the L4 synchronized worms were cultured at 20 °C for 24 h. The samples (0.4 mg mL −1 and 0.8 mg mL −1 ) with 5-fluorouracil were added to the NGMs, and the worms were further cultured for 48 h. Then, the worms were transferred to 96-well plates with methyl viologen (200 μL, 70 mmol L −1 ) or H 2 O 2 (200 μL, 4 mmol L −1 ) and cultured at 20 °C. The number of dead worms was recorded every 60 min.…”
