Global analysis of hematopoietic and vascular endothelial gene expression by tissue specific microarray profiling in zebrafish

Abstract: In this study, we utilize fluorescent activated cell sorting (FACS) of cells from transgenic zebrafish coupled with microarray analysis to globally analyze expression of cell type specific genes. We find that it is possible to isolate cell populations from Tg(fli1:egfp)(y1) zebrafish embryos that are enriched in vascular, hematopoietic and pharyngeal arch cell types. Microarray analysis of GFP+ versus GFP- cells isolated from Tg(fli1:egfp)(y1) embryos identifies genes expressed in hematopoietic, vascular and p… Show more

“…Embryo dissociation and FACS were performed as described previously by Covassin et al [44]. The GFP + and GFP − cells were separately collected from cmyb:EGFP and runx1:EGFP transgenic embryos (Zhang P and Liu F, unpublished data) using MoFlo XDP (Beckman).…”

“…Larval dissociation was performed as described (Covassin et al, 2006). Briefly, larvae were killed in a bath of ice-cold E3 and rinsed in calcium-free Ringer solution before trypsin (Invitrogen, final concentration of 0.25% trypsin-EDTA [ethylenediaminetetraacetic protein]) digestion.…”

“…They achieved this by expressing EGFP under the promoter control of a gene, which was uniquely expressed in the desired cell population. [44][45][46][47][48] Another significant drawback to defining cells of interest by their endogenous cell surface markers is that these markers may not be expressed at the earliest stages of differentiation. Onset of stem cell differentiation down a particular lineage is often defined by a transcription factor functioning as a master regulator.…”
