Global Distribution of the Reniform Nematode Genus Rotylenchulus with the Synonymy of Rotylenchulus macrosoma with Rotylenchulus borealis

Abstract: Reniform nematodes of the genus Rotylenchulus are semi-endoparasites of numerous herbaceous and woody plant roots that occur largely in regions with temperate, subtropical, and tropical climates. In this study, we compared 12 populations of Rotylenchulusborealis and 16 populations of Rotylenchulusmacrosoma, including paratypes deposited in nematode collections, confirming that morphological characters between both nematode species do not support their separation. In addition, analysis of molecular markers usin… Show more

“…For molecular analyses, and to avoid mistakes in case of mixed populations in the same sample, single nematodes were previously mounted in a drop of NaCl and used for molecular identification after recording morphological data. Genomic DNA extraction from single specimens was conducted as described by Palomares-Rius et al [ 68 ]. Briefly, an individual nematode is cut using a scalpel in a drop of PCR buffer (ThermoPol ® , Biolabs, New England, USA) (20 µL) and 2 μL proteinase K (600 μg/mL) were added.…”

“…All necessary morphological and morphometrical data by taking pictures and measurements using the above camera-equipped microscope were recorded. Then DNA extraction from single individuals was performed as described by Palomares-Rius et al [ 57 ], and more importantly, for all the 24 studied isolates, all the three molecular markers of each Paratylenchus isolate belong to the same single extracted individual in each PCR tube without any exception. In addition, male and juveniles conspecificity was proven by single DNA extraction of male or juveniles for each species.…”

“…DNA extraction was always based on single nematode specimens as defined by Palomares-Rius et al [ 54 ], and more decisive, for all the 27 considered populations, all the three molecular markers of each Paratylenchus population are coming from the same single DNA extracted nematode in individually PCR tube without any exemption. Furthermore, assignation of male and juvenile stages to one species always was proven by single DNA extraction of these individuals.…”
