Globin synthesis and erythroid differentiation in a Friend cell variant deficient in heme synthesis.

Abstract: Friend erythroleukemia cells of line Fw are noninducible for hemoglobin synthesis by dimethyl sulfoxide, butyric acid, and other agents. However, these agents were found to induce hemoglobin synthesis if the cells were also treated with exogenous hemin. Butyric acid (or, to a lesser extent, dimethyl sulfoxide) by itself induced accumulation of the erythroid-specific membrane protein spectrin. The basis of the control of globin gene expression by hemin was investigated. Hemin does not control globin and total p… Show more

“…Besides its role in translational control in reticulocytes (1)(2)(3) it also participates in the induction of globin mRNA and in erythroid differentiation (14)(15)(16)(17)(18). Recently it was reported that hemin enhances the differentiation of mouse 3T3 cells to adipocytes (19).…”

“…2B ) obtained when the MEL cells are treated with 2% and 2.5 mM DMSO and HMBA, respectively. The high level of induction (>80% after 3 or 4 days in all the experiments performed) confirms that this cellular system is excellent to determine inhibitors of the expression of adult β-globin genes, since, unlike other erythroid cellular model systems (such as human K562 cells), these cells mostly produce the adult-type Hb major and Hb minor hemoglobins ( 39 ). As shown in Fig.…”

“…MEL cells ( 37 39 ) were cultured in humidified atmosphere of 5% CO 2 /air in RPMI-1640 medium (Sigma-Aldrich) supplemented with 10% fetal bovine serum (FBS; Biowest, Nuaillé, France), 50 U/ml penicillin and 50 μg/ml streptomycin ( 39 ). DMSO and HMBA were from Sigma-Aldrich.…”

“…Under these conditions endogenous hemin is probably not synthesized. Indeed, exogenous hemin has been demonstrated to inhibit heme synthesis in K-562 cells (Bottomley et al, 1985) and to support Hb production in MEL clones genetically deficient in heme synthesis (Rutherford and Harrison, 1979).…”
