Glucagon activates mitochondrial 3‐hydroxy‐3‐methylglutaryl‐CoA synthase in vivo by decreasing the extent of succinylation of the enzyme

Abstract: 1. 3-Hydroxy-3-methylglutaryl-CoA (HMG-CoA) synthase (EC 4.1.3.5) in extracts of rat liver mitochondria can be inactivated by succinyl-CoA and activated by incubation in a medium designed to cause desuccinylation ('desuccinylation medium'). 2. The enzyme is less active in extracts of whole liver from control rats than from rats treated with glucagon or mannoheptulose. Incubation in desuccinylation medium raises the activity in extracts from control rats to the same value as treated rats, suggesting that the ex… Show more

“…The factors responsible for the postnatal and starvation-associated induction of HMGCS2 gene expression have been identified and shown to involve changes in circulating hormone and fatty acid levels. Administration of dibutyryl-cyclic AMP to adult rats increases mRNA, protein levels, and enzymatic activity of HMGCS2 in the liver (6,9). Similar effects are produced by a high-fat diet (10), whereas insulin causes the opposite effect (6).…”

“…Administration of dexamethasone or dibutyryl-cAMP to adult rats elicits increased liver mRNA, protein and enzymic activity [7,8,16], whereas opposite results are observed with insulin [7]. In addition, the rat mitochondrial HMG-CoA synthase gene contains elements that mediate its multihormonal regulation and tissue specificity [17].…”

“…Activity of HMGCS2, the enzyme-regulating flux through ketogenesis, is decreased by acetylation and succinylation, and increased by phosphorylation (Quant et al 1990, Shimazu et al 2010, Grimsrud et al 2012. HMGCS2 is phosphorylated by PKA, a downstream glucagon signaling molecule, and hyperphosphorylation of HMGCS2 occurs in obese rodents (Grimsrud et al 2012).…”
