Glucocorticoid Regulation of Bone Sialoprotein (BSP) Gene Expression. Identification of a Glucocorticoid Response Element in the Bone Sialoprotein Gene Promoter

Abstract: Glucocorticoids modulate the development and growth of many organs through interactions with a specific intracellular receptor (glucocorticoid receptor) that regulates gene transcription through a cognate element, the glucocorticoid response element (GRE), in the promoter of target genes. In bone formation glucocorticoids stimulate osteoblast differentiation and the formation of bone matrix. Recent studies have demonstrated that the induction of the bone sialoprotein (BSP) gene is associated with osteoblast di… Show more

“…Glucocorticoids, including Dex, modulate the development and growth of many organs through interactions with a specific intracellular receptor (glucocorticoid receptor) that regulates gene transcription through a cognate element, the glucocorticoid response element (GRE), in the promoter of target genes (Ogata et al 1995). We searched about GRE in TASK-1 and TWIK-2 gene by using NCBI (National Center for Biotechnology Information) data base, but were not able to find the representative nucleotide sequence (AGAACA nnn TGTTCT) corresponding to the response element.…”

“…Both proteins belong to the small integrin-binding ligand N-linked glycoprotein (SIBLING) family and are structurally related [11]. The runt-related transcription factor 2 (Runx2) and transforming growth factor b (TGF-b) are involved in the regulation of their gene expression [12][13][14]. Both OPN and BSP contain an Arg-Gly-Asp (RGD) motif, which mediates binding to surface proteins of tumour and normal cells, such as the integrin a v b 3 receptor [11].…”

“…expression in a PKA-dependent way (McCarthy et al, 2000a). This may account, at least in part, for a permissive, and in some cases critical role for glucocorticoid on bone matrix protein synthesis (Raisz and Chyun, 1984;Kimoto et al, 1994;Ogata et al, 1995).…”
