Glucose Limitation Sensitizes Cancer Cells to Selenite-Induced Cytotoxicity via SLC7A11-Mediated Redox Collapse

Abstract: Combination of intermittent fasting and chemotherapy has been drawn an increasing attention because of the encouraging efficacy. In this study, we evaluated the anti-cancer effect of combination of glucose limitation and selenite (Se), a representative inorganic form of selenium, that is preferentially accumulated in tumors. Results showed that cytotoxic effect of selenite on cancer cells, but not on normal cells, was significantly enhanced in response to the combination of selenite and glucose limitation. Fur… Show more

“…Our results revealed that a high level of selenium alleviated the oxidized fish oil-induced reduction of the GSH content and slc7a11 expression and alleviated the oxidized fish oil-induced increase of atf3 mRNA expression. Studies have suggested that oxidative stress increased the ATF3 expression [ 47 ], and that selenite up-regulated the expression of SLC7A11 [ 48 ]. Thus, it is reasonable to speculate that ATF3 mediated the alteration of slc7a11 mRNA expression by the use of selenium and oxidized fish oils, thus, this affected the intestinal GSH content.…”

“…Selenium (Se) is an essential trace element for humans and animals, plays pivotal roles in antioxidant defense, anti-cancer and detoxification [155][156][157] . The selenium-containing enzyme GPX4 is currently recognized as a central repressor of ferroptosis, thus it is usually considered as ferroptosis inhibitor [48,158] .…”

“…BAY-876 manifests antineoplastic action mainly via its ability to inhibit GLUT1 leading to multiple actions on neoplastic cells, including suppression of proliferation, drug-resistance, EMT, and metabolism ( Yang et al, 2021 ). BAY-876 was reported to manifest increased NADP + /NADPH simulating glucose starvation conditions ( Chen et al, 2022 ). However, the antineoplastic activity of BAY-876 needs to be validated on a broader range of neoplastic cells.…”
