Glutathione and gamma-glutamyl cycle enzymes in crypt and villus tip cells of rat jejunal mucosa.

Abstract: Villus tip cells and crypt cells of rat jejunal mucosa were separated bythe planing procedure of Imondi et a]. and were studied with respect to their activities of the enzymes of the 'y-glutamyl cycle and glutathione content. The villus tip cells exhibit much higher y-glutamyl transpeptidase activities than do the crypt cells; thus, y-glutamyl transpeptidase appears to be a villus-specific enzyme. y-Glutamyl cyclotransferase and the enzymes required for glutathione synthesis are not specifically localized to … Show more

“…The origin of both the enteroendocrine and intestinal epithelial cells is believed to arise from a primitive intestinal stem cell located in the intestinal crypt layers (31). Published studies have shown that two transcription factors, cdx-2 and Pdx-1, may be important in deciding the pathway leading to differentiation and thus the formation of absorptive enterocytes or pancreatic cells, respectively (32).…”

“…Not surprisingly, during this process the epithelial cells undergo extensive biochemical, functional and morphological alterations [5,7,10,25,27,28,30,35] which include changes in tight junction structure [18]. While tight junctions of villus absorptive cells are rather uniform in structure, those between undifferentiated crypt cells are more variable and, on the average, have fewer strands and are less deep than those between villus absorptive cells [18].…”

“…One of such non-enzymatic antioxidants is a glutathione (GSH). GSH is a tripeptide thiol synthesized by glutamic acid, cysteine and glycine in the pathway of γ-glutamyl cycle [77,78] and is present in either reduced (GSH) or oxidized (GSSG) form. GSH acts as a strong reducing agent as well as serving an electron donor for glutathione peroxidase (Fig.…”
