Gluten Determination by Gliadin Enzyme-Linked Immunosorbent Assay Kit: Interlaboratory Study

Abstract: An interlaboratory study with 10 participants was performed to obtain validation and performance data for an enzyme-linked immunosorbent assay (ELISA) kit developed for quantitative gluten determination in foods. The ELISA kit used for this study is based on 2 monoclonal and 1 polyclonal antibody developed by Immunotech, a Beckman Coulter Co. This kit did not show any false positive results or cross-reactivity with oat, rice, maize, and buckwheat. The gliadin standard from the Working Group on Prolamin Analysi… Show more

“…The purified gliadin standard developed by the WGPAT ( van Eckert et al, 2006 ) has been used by European laboratories for research on gluten ( Dahinden, von Büren, & Lüthy, 2001 ; Gabrovská et al, 2006 ; Köppel, Stadler, Lüthy, & Huèbner, 1998 ; van Eckert et al, 2010 ). The WGPAT standard consists of a gliadin extract from 28 European wheat cultivars of the 1999 harvest.…”

“…At least in sandwich form, the R5-ELISA is considered more sensitive than the anti-ω-gliadin ELISA and better at detecting barley gluten [ 2 , 24 ]. An inability to detect barley and rye sequences is a weakness of another recently-developed commercial gluten ELISA [ 6 , 25 ].…”

“…New improved gluten ELISAs have since appeared on the market [ 5 , 6 ] and others are in development [ 7 , 8 ]. The commercial R-ELISA [ 5 ] has been deemed sufficiently reliable and sensitive to support standards for GF foods based on final gluten content [ 9 ].…”

Traditionally, the calibration of an analytical instrument has been achieved through an ordinary least-squares linear regression (OLR) model.…”

“…The discordancy tests (single-as well as multipleoutlier types) originally proposed by both Dixon (1951) and Grubbs (1950Grubbs ( , 1969 have been very popular and are still in wide use in different fields (e.g., Barnett and Lewis 1994, Li et al 2003, Serbst et al 2003, Farre et al 2006, Gabrovská et al 2006, Sang et al 2006, Verma and Quiroz-Ruiz 2006a, b, 2008, Hayes et al 2007. Several other discordancy tests are also available for this purpose, most notable among which are the skewness and kurtosis tests (Barnett and Lewis 1994, Verma 1997, Velasco and Verma 1998, Verma and Quiroz-Ruiz 2006a, b, 2008.…”
