Glycosylated Cathepsin V Serves as a Prognostic Marker in Lung Cancer

Abstract: Dysregulation of cysteine cathepsin protease activity is pivotal in tumorigenic transformation. However, the role of cathepsin protease in lung cancer remains unknown. Here, we analyzed GEO database and found that lung cancer presented high expression of cathepsin V (CTSV). We then performed immunohistochemistry assay in 73 paired lung cancer tissues and normal lung tissues and confirmed that CTSV is overexpressed in lung cancer and correlates with poor prognosis. The mass spectrometry experiment showed that t… Show more

“…Researches indicate that abnormal proteolytic activity may trigger the manifestation of diseased conditions [18]. CTSV had been proved to be a proteolytic enzyme that is overexpressed in metastasis lung cancer patients and predicted poorer outcome [19], here we speculated that CTSV interact with certain substrates and remodel ECM environment. To this end, we performed protein mass spectrometry of both NCI-H1975 and A549 cells and identi ed that CTSV interacts with bronectin, E-cadherin and Ncadherin, which are essential molecules for metastasis (Supplementary information, Figure S2a).…”

“…Despite its involvement in various cancers, the specific role of CTSV in cSCC remains unclear. Notably, high CTSV expression has been linked to unfavourable outcomes in breast ductal carcinoma, 33 promotes proliferation through the NF‐κB pathway in bladder cancer, 34 and drives lung cancer metastasis 35 . Conversely, CTSV is reported to be decreased in melanoma samples 36 .…”

“…Notably, high CTSV expression has been linked to unfavourable outcomes in breast ductal carcinoma, 33 promotes proliferation through the NF-κB pathway in bladder cancer, 34 and drives lung cancer metastasis. 35 Conversely, CTSV is reported to be decreased in melanoma samples. 36 To fully comprehend the role of CTSV in cSCC development, further investigation is warranted.…”

“…The advent of high-resolution MS has led to the discovery of novel, specifically expressed protein biomarkers that may be involved in the pathophysiology of diseases that are difficult to identify by immunohistochemistry and histologic morphology alone [ 66 ]. The importance of MS in identifying the target proteins involved in virtually any disease process makes it an invaluable discovery tool since MS can provide a catalog of novel target proteins implicated in health and disease.…”
