GM-CSF Production by Tumor Cells Is Associated with Improved Survival in Colorectal Cancer

Abstract: H-thymidine incorporation. Expression of cytokine genes in colorectal cancer and autologous healthy mucosa was tested by quantitative, real-time PCR. A tumor microarray (TMA) including >1,200 colorectal cancer specimens was stained with GM-CSF-and M-CSF-specific antibodies. Clinicopathological features and overall survival were analyzed. Results: GM-CSF induced CD16 expression in 66% AE 8% of monocytes, as compared with 28% AE 1% in cells stimulated by M-CSF (P ¼ 0.011). GM-CSF but not M-CSF-stimulated macropha… Show more

“…In addition, combination of GM-CSF vaccination with radiotherapy in patients with metastatic solid tumors in a recent clinical trial, resulted in increased treatment response, by presumably enhancing the dendritic cell cross-talk, and overcoming the immunosuppressive effect of radiation [43]. The above observation is in line with another recent publication in colon cancer, where higher tissue levels of GM-CSF at the tumor cells was correlated with lower cancer stage and prolonged survival [44]. Although the exact mechanism associated with these observed changes remains unknown, based on the presented data, we may safely assume an involvement of the non-canonical Wnt/Ca 2+ signalling.…”

“…It is the critical molecular in inducing the TAMs polarization to M1 phenotype. The production of GM‐CSF by tumor cells is an independent favorable prognostic factor in colon cancer . IL12 is a heterodimer that is composed of two subunits, IL12α and IL12β with the active form of IL12 p70 .…”

“…More recently, this has been used in the treatment of a wide range of malignancies [13]. Nebiker et al [11] further unraveled an important paradoxical colorectal cancer feature, which is represented by the favorable prognostic role of GM-CSF. Immune-dependent and immune-independent antitumor activities of GM-CSF in human colorectal cancer have been suggested.…”

“…With the use of dose-intensive chemotherapy, GM-CSF has been widely applied to minimize chemotherapy-induced myelosuppression. In addition, GM-CSF has also been specifically used as an anti-tumor agent with varying degrees of success [11,12]. The expression of GM-CSF and its receptor genes within human glioma specimens have been previously reported.…”
