GNA11 differentially mediates fibroblast growth factor 2‐ and vascular endothelial growth factor A‐induced cellular responses in human fetoplacental endothelial cells

Abstract: During pregnancy, fetoplacental angiogenesis is dramatically increased in association with rapidly elevated blood flow. Any disruption of fetoplacental angiogenesis may lead to pregnancy complications such as intrauterine growth restriction. Fibroblast growth factor 2 (FGF2) and vascular endothelial growth factor A (VEGFA) are crucial regulators of fetoplacental angiogenesis. G protein α subunits q (GNAq) and 11 (GNA11) are two members of the Gα subfamily involved in mediating vascular growth and basal blood p… Show more

“…It is noteworthy that although we have verified the specificity of Ad‐ GNA14 (Zou et al, ), Ad‐ GNA14 at high doses (10 and 20 MOI) elevates GNA11 protein levels in HUVECs. It is unclear if Ad‐ GNA14 also alters expression of other Gα q/11 subfamily members such as GNAq as they all are high identical in amino acid sequences.…”

“…On the other hand, it was ultimately not surprising to us that VEGFA treatment alone did not fully rescue FGRadv EC migratory defects because ARNT has other downstream targets other than VEGFA. Moreover, others have found that ablation of key mediators of cardiovascular function, such as the G proteins G αq or G α11 , inhibit VEGFA-induced human umbilical vein EC (HUVEC) proliferation and migration, suggesting that even if VEGFA is present in adequate quantities, other pathways are also important in mediating EC migration and angiogenesis (50)(51)(52). Likewise, it would also be extremely unlikely for an organ as complex as the human placenta to be regulated by just one angiogenic growth factor.…”
