Gold-nanorod contrast-enhanced photoacoustic micro-imaging of focused-ultrasound induced blood-brain-barrier opening in a rat model

Abstract: Abstract. In this study, we develop a novel photoacoustic imaging technique based on gold nanorods (AuNRs) for quantitatively monitoring focused-ultrasound (FUS) induced blood-brain barrier (BBB) opening in a rat model in vivo. This study takes advantage of the strong near-infrared absorption (peak at ∼800 nm) of AuNRs and the extravasation tendency from BBB opening foci due to their nano-scale size to passively label the BBB disruption area. Experimental results show that AuNR contrast-enhanced photoacoustic … Show more

“…In addition to active transport, the BBB can also be chemically or physically opened to allow the free diffusion of contrast agents [90]. Li et al used IRDye800-c(KRGDf) to target the overexpressed integrin α v β 3 in brain glioblastoma [61].…”

“…In the case of NRs, the two characteristic SPR modes are transverse and longitudinal SPR or normal and parallel to the long axes of the NRs, respectively, and take place at specific wavelength bands, called plasmon bands [1][2] . For noble metals, the plasmon bands are positioned in the UV -Visible range, features that are advantageous in numerous applications [3][4][5][6][7][8][9] .…”

“…Other methods besides MRI have been used to detect FUS-induced BBB opening, such as single photon emission computed tomography (SPECT) with i.v. administration of 99m TC-DTPA radiotracer as the contrast agent, or small-animal photoacoustic imaging with gold nanorods 15, 16. In addition, dynamic changes in microvasculature during FUS-induced BBB opening have been microscopically observed by fluorescent multi-photon microscopy, and with a high-speed camera 17, 18.…”
