Golgi/granule processing of peptide hormone and neuropeptide precursors: A minireview

Abstract: Proteolytic processing of precursor proteins is a phylogenetically ancient and widely used mechanism for producing biologically active peptides. Proteolytic cleavage of proproteins begins only after transport to the Golgi apparatus has been completed and in most systems may continue for many hours within newly formed secretory vesicles as these are stored in the cytosol or transported along axons to more peripheral sites of release. Paired basic residues are required for efficient proteolysis in most precursor… Show more

“…The IM domain of pro-SI contains a Ser/Thr-rich domain, the site of heavy O-glycosylation within the pro-SI species. This type of glycosylation commences in the cis-Golgi and is terminated in the trans-Golgi network (31). Because IM is not O-glycosylated and persists exclusively as an endo Hsensitive polypeptide the data demonstrate that this form did not traverse the ER to the Golgi apparatus to be processed to a complex glycosylated protein.…”

“…Finally, ICA69 overlapped in part with proinsulin, which is found in ISGs, but not in MSGs (Fig. 3E) (45,46). It also co-distributed with insulin in the Golgi region but not near the plasma membrane (Fig.…”
