Gomisin N inhibits adipogenesis and prevents high-fat diet-induced obesity

Abstract: Gomisin N (GN) is a physiological lignan derived from Schisandra chinensis. In the present study, we investigated the inhibitory effects of GN on differentiation of 3T3-L1 preadipocytes and the anti-obesity effects of GN in high-fat diet (HFD)-induced obese mice. Incubation with GN significantly inhibited the differentiation of 3T3-L1 preadipocytes in a dose-dependent manner. This inhibitory effect primarily occurred at an early adipogenic stage through impairment of mitotic clonal expansion (MCE) caused by ce… Show more

“…In addition, ESEE reduced HFd-induced serum markers of fatty liver, including AST and ALT. These findings were similar to those reported for the anti-obesity effect of Gomisin N in HFd-induced obese mice (26). ESEE reduced HFD-induced increases in serum lipid profiles, such as TC, TG and LdLc, and increased serum levels of HdLc, which were in accordance with its observed suppressive effect on HFd-induced fatty liver and hyperlipidemia in HFd-fed mice.…”

“…Another approach would be to limit adipogenesis by inhibiting both the proliferation and differentiation of adipocytes. Plant extracts have been screened to identify compounds that can suppress adipogenesis [8]. One such plant extract that has attracted much interest is Oroxylum indicum.…”

“…Among them, 3T3-L1 cells were presented by Howard Green in 1974 and are commonly used as a cell model to study differentiation in adipocytes [7]. Differentiation from preadipocytes to adipocytes consists of three stages: growth arrest, increase by somatic cell replication, and differentiation [8]. The cell cycle of preadipocytes, which have ceased growth by cell conjugation, is activated by cell differentiation-inducing media containing IBMX (3-isobutyl-1-methylxanthine), dexamethasone, and insulin.…”
