GouR, a TetR Family Transcriptional Regulator, Coordinates the Biosynthesis and Export of Gougerotin in Streptomyces graminearus

Abstract: Gougerotin is a peptidyl nucleoside antibiotic. It functions as a specific inhibitor of protein synthesis by binding ribosomal peptidyl transferase and exhibits a broad spectrum of biological activities. gouR, situated in the gougerotin biosynthetic gene cluster, encodes a TetR family transcriptional regulatory protein. Gene disruption and genetic complementation revealed that gouR plays an important role in the biosynthesis of gougerotin. Transcriptional analysis suggested that GouR represses the transcriptio… Show more

“…As a large and important family of one-component signal transduction systems, TFRs are widely associated with metabolism, antibiotic production, quorum sensing, and multidrug resistance (15,27). In the present study, we discovered that SLCG_2919 can regulate lincomycin biosynthesis by repressing the transcription of lincomycin biosynthetic structural genes, resistance genes, and a regulatory gene in the lin cluster, as well as by repressing its adjacent gene, SLCG_2920, which encodes an ATP/GTP binding protein.…”

“…It is noteworthy that the PCC activity of the DBX(pccBX) strain was lower than that of the DF50 strain. The phenomenon of partial complementation of mutant phenotypes is widely found in different organisms (28). In the complemented DBX(pccBX) strain, the pccBX genes under the control of their native promoter were carried on a plasmid instead of being integrated into their original sites in the genome.…”

“…Mfo1, a TetR family regulator, was found to negatively regulate the expression of mfs1 , which is consistent with previous observations that members of the TetR family of regulators can repress genes whose products are involved in multidrug resistance (Ramos et al ., ). In most cases, binding of TetR to its cognate DNA sequence could be dissociated by small‐molecule ligands, including antibiotic end products and their biosynthetic intermediates (Zhang et al ., ; Wei et al ., ). For instance, in biosynthesis of simocyclinone D8, the intermediate, simocyclinone C4, is effective in SimR dissociation in the regulation of the efflux pump gene simX (Le et al ., ).…”
