Granule Localization of Glutaminase in Human Neutrophils and the Consequence of Glutamine Utilization for Neutrophil Activity

Abstract: The provision of glutamine in vivo has been observed to reduce to normal levels the neutrophilia observed after exhaustive exercise and to decrease the neutrophil chemoattractant, interleukin-8. Thus, the role for glutamine in the regulation of inflammatory mediators of human neutrophil activation was investigated. The study sought to establish whether glutamine supplementation in vitro affects neutrophil function at rest and whether glutaminase, the major enzyme that metabolizes glutamine, is present in human… Show more

“…The underlying thesis is, that in anabolic situations not only a raised pyruvate uptake, but also a substantial amount of de novo intracellular pyruvate as well as pyruvate-dependent metabolite synthesis occurs [37]- [39]. And indeed, our results seem to confirm this hypothesis, because following pyruvate incubation increased glutamate, glutamine, aspartate (asparagine), alanine, α-ketotglutarate but also to serine and glycine concentrations have intracellularly been found (i.e.…”

“…Indeed, GLS2 has also been detected in brain, 5 pancreas, 6 cancer cells, 7 and cells of the immune system. 8 Both isoforms have been identified in human and mouse brain. At the subcellular level, GLS2 localizes to the inner mitochondrial membrane, while in neurons it has been observed in the nucleus.…”

“…Several ammonia-forming pathways may be active in a brain abscess, including ammonia formation from glutamine by phosphate-activated glutaminase, an enzyme that may originate from invading leukocytes 5 or from damaged neurons. 13 In the present study, S. intermedius and S. aureus produced ammonia at high concentrations in vitro, but seemed to do so along somewhat different pathways.…”
