Green Synthetic Approach for Photo-Cross-Linkable Methacryloyl Hyaluronic Acid with a Tailored Substitution Degree

Abstract: The conventional synthesis of methacryloyl hyaluronic acid (HA-MA) requires an extremely high amount of modification reagents, the organic solvents, and strenuous purification steps. Herein, a new green synthetic approach for the methacryloyl hyaluronic acid preparation with a tailorable substitution degree (SD) is reported, in which methacryloyl hydrazide is used as a more reactive reagent and only water is used as the solvent. The new method significantly reduces the amount of functionalization reagents (as … Show more

“…Characterization of PDA NPs@HAMA Nuclear magnetic resonance spectroscopy (1H NMR) results revealed that the emerging signals at 5.6 and 6.2 ppm in HAMA, belonging to the protons of -CH=CH2, which confirmed that methacrylic acid was successfully connected to the skeleton of HA (Figure 1a). 30 Compared with HA, the stretching vibration band of C=O in HAMA was divided into two peaks, demonstrating the presence of anhydride (Figure 1b). The enhanced C-O-C stretching vibration also indicated that the ester bond was formed after modification, further verifying the conjugation of the carbon-carbon double bond in HAMA that endowed the cross-linkable and photocurable functions.…”

“…Methacryloyl hydrazide was synthesized following a reported procedure, which was slightly modified . Methacrylic anhydride (10.35 g, 0.067 mol) was dissolved in chloroform (40 mL) and then added dropwise to hydrazine monohydrate in a 50 mL 2-necked round bottom flask (14 mL, 0.288 mol) at 0 °C for 30 min.…”

“…In the realm of tissue engineering, hyaluronic acid has been increasingly utilized as a scaffolding biopolymer. Islet cells that were encapsulated in hyaluronic acid and alginate matrices have reportedly shown high viability and insulin secretion [160]. Additionally, hyaluronic acid-alginate hybrid microcapsules have been demonstrated by Cañibano-Hernández et al [158] to decrease apoptosis and boost β-cell survival.…”
