Griffipavixanthone, a dimeric xanthone extracted from edible plants, inhibits tumor metastasis and proliferation via downregulation of the RAF pathway in esophageal cancer

Abstract: Metastasis causes a large number of deaths among esophageal cancer patients. The activation of RAF family proteins elevates tumor metastasis and proliferation. In screen targeting the RAF protein, we identified that Griffipavixanthone (GPX), a dimeric xanthone isolated from Garcinia esculenta, is a B-RAF and C-RAF inhibitor against esophageal cancer cells. Using wound healing, transwell migration and matrigel invasion assays, we confirmed that GPX significantly inhibited cell migration and invasion. Furthermor… Show more

“…Our previous studies showed that compounds isolated from Garcinia species had potent inhibitory effect on esophageal cancer cell metastasis 10 18 . In addition, the EtOAc-soluble fraction of the actone extract of G. oblongifolia leaves could inhibit TE1 cell migration at concentrations of 12.5, 25 and 50 μg/ml ( Figure S20 ).…”

“…Next, we evaluated the constitutive activation status of STAT3, FAK, and p44/42 MAPK. Similar to STAT3, FAK and p44/42 MAPK signaling pathways are also involved in the proliferation of esophageal cancer . As shown in Figure b, STAT3 was constitutively phosphorylated at high levels in seven ESCC cell lines (TE1, TE4, TE5, TE6, TE8, TE10, and TE15 cells).…”

In contrast, p-STAT3 levels in three ESCC cell lines (TE9, TE11, and TE14) were low.…”
