Growth-inhibitory effects of the chemopreventive agent indole-3-carbinol are increased in combination with the polyamine putrescine in the SW480 colon tumour cell line

Abstract: Background: Many tumours undergo disregulation of polyamine homeostasis and upregulation of ornithine decarboxylase (ODC) activity, which can promote carcinogenesis. In animal models of colon carcinogenesis, inhibition of ODC activity by difluoromethylornithine (DFMO) has been shown to reduce the number and size of colon adenomas and carcinomas. Indole-3-carbinol (I3C) has shown promising chemopreventive activity against a range of human tumour cell types, but little is known about the effect of this agent on … Show more

“…These indole derivatives have been shown to suppress the proliferation of various cancer cell lines at the concentration range between 1 and 100 mM. These include those of breast [14][15][16][17], colon [18][19][20], and prostate [21][22][23], by targeting a wide spectrum of signaling pathways governing hormonal homeostasis, cell-cycle progression, and cell proliferation and survival [24][25][26][27][28]. Previous studies have also shown DIM and I3C can suppress the proliferation of prostate cancer cells [29][30][31].…”

“…I3C has been shown to suppress the proliferation of a wide variety of cells, including breast cancer cells, [9][10][11][12] colon cancer cells, [13][14][15] prostate cancer cells, 16,17 and endometrial cancer cells. 18 Katdare's group showed that I3C inhibits benzopyrene-induced proliferation of mammary epithelial cells.…”

“…In addition to Mig-7 mRNA being produced under the CMV promoter in this cell line, ODC activity is a log higher than those found in other carcinoma cell lines. As a result, HT29 cells are frequently used to study ODC activities (Chi et al, 2006;Hudson et al, 2003;Martinez et al, 2003;Selamnia et al, 1998). Parental HT29 cells do not express Mig-7 mRNA due to a lack of signaling from αvβ5 integrin (Crouch et al, 2004).…”
