Growth of malignant extracranial tumors alters microRNAome in the prefrontal cortex of TumorGraft mice

Abstract: A wide array of central nervous system complications, neurological deficits, and cognitive impairments occur and persist as a result of systemic cancer and cancer treatments. This condition is known as chemo brain and it affects over half of cancer survivors. Recent studies reported that cognitive impairments manifest before chemotherapy and are much broader than chemo brain alone, thereby adding in tumor brain as a component. The molecular mechanisms of chemo brain are under-investigated, and the mechanisms o… Show more

“…Immunodeficient female mice (nu/nu athymic) were subcutaneously injected with the fresh specimen of the tumor (TumorGraft™) into the flanks, followed by the treatment with DOX/PTX/CPP. The results showed the deregulation of different miRNA families, especially miR-200 and miR-183/96/182 clusters in tumor-bearing and chemotherapytreated animals (Kovalchuk et al 2017). Dysregulation of miRNA leads to alteration in the levels of BDNF, which plays a vital role in cognition and memory (Baydyuk and Xu 2014).…”

“…Homozygous mice exposed to chemotherapeutic agents show more pronounced disruptions in post-transcriptional regulation of gene expression, mainly miRNA changes in the prefrontal cortices. miRNA dysregulation is associated with the altered levels of brain-derived neurotrophic factor (BDNF) that plays a key role in cognition and memory ( 71 ).…”

“…It could provide a large amount of data, and could be useful to predict the roles of individual miRNAs and their pathways. Nowadays, the miRNAomes of different mammal tissues have been revealed by the next-generation sequencing, including mice [8], cattle [9], and pig [10]. However, the study of miRNAome in Large White livers of different developmental stages is still lacking.…”
