In vitro: regeneration of Chenopodium quinoa Willd. was achieved from cotyledonary nodes explants. In this study, used 6-Benzylaminopurine (BAP) and α-Naphthalene Acetic Acid (NAA) of plant growth regulators with different concentrations individually as well as in combination and used different concentrations of sugar (sucrose) with different concentrations. For was rooting, used half strength (½MS), full-strength MS and ½ MS supplemented with 0.2 mg/l of NAA. The results mentioned, explant responding (%) to multiplication was about 73% for all BAP treatments compared with control and average numbers of shoot increased with increased BAP concentration except 5 mg/l of BAP. The highest explant responding (%) was in media supplemented BAP without NAA compared other treatments noted that the media with combination of BAP and NAA gives formation of callus in bases of the plantlets. Also, the result inducted the combinations between (BAP–NAA) was highly significantly (P≤ 0.001) and less effective on number of shoots where the highest number of shoot was 3.40 in media with 3 mg/l BAP compared other treatments. The highest of explant responding 93.33% was in media supplement with 10 g/l sucrose and (10 g/l sucrose + 3 mg/l BAP), but sucrose level for good greening and developed shoots (4 shoots) was in medium supplement with 10 g/l sucrose. The shoots rooted well on half-strength MS medium with 60% percentage of root. The rooted shoots were acclimatized and transferred to green house to follow their development.

ÖZET: Bu çalışma, bitki hormonlarından (BH) naftalen asetik asit (NAA), 6-benzil amino pürin (BAP), kinetin (KIN) ve thidiazuron (TDZ)'un korunga (Onobrychis sativa L.) bitkisinin bazı in vitro özelliklerine olan etkilerini belirlemek amacıyla yapılmıştır. İki farklı deneme şeklinde yürütülen ve toplam 42 farklı yöntemin denendiği bu çalışma iki aşamada gerçekleştirilmiştir. Birinci aşamada tohumlar hormonsuz ortamda çimlendirilmiş, ikinci aşamada ise hormonsuz ortamda çimlendirilmiş tohumlardan meydana gelen bitkiciklerden eksplantlar (kotiledon, kök ve hipokotil) alınarak farklı hormon, doz ve hormon-doz kombinasyonlarını içeren ortamlara aktarılmıştır. Bitki hormonlarının etkisine bakıldığında en fazla sayıda sürgün oluşumu kök eksplantından 29.0 adet ile 1.5 mg l -1 BAP'dan doksanıncı gün sonunda elde edilmiştir. Kotiledonlarda kültür süresindeki artışa bağlı olarak sürgün sayısı artmış ve en fazla sürgün 7.8 adet ile TDZ'nin 1.0 mg l -1 'lik dozundan doksanıncı günün sonunda elde edilmiştir. Kök oluşumu bakımından en iyi sonuç ise 14.8 adet ile 0.4 mg l -1 NAA'dan hipokotilden elde edilmiştir.Anahtar Kelimeler: Bitki hormonları, doku kültürü, korunga, Onobrychis sativa L.ABSTRACT: This study prepared was carried out determining plant hormones (PH) (NAA, BAP, KIN and TDZ) on formation of tissue culture on the sainfoin plants (Onobrychis sativa L.) grown in in-vitro conditions. This study which is applied 42 different methods was performed in two sections. In the first stage, the seeds were sprouted in hormone-free environment. In the second stage, the explants (cotyledon, root and hypocotyl) obtained from the plantlets were transferred to the mediums including different hormone, dose and hormone-dose combinations. For the effect of plant growth hormones, the highest number of shoots (29.0 nr) was obtained from root explant the dose 1.5 mg l -1 of BAP in the end of 90th day. Shoot number on cotyledones increased depending on incereasing number of treatment/day and the highest number of shoot (7.8 nr) was obtained from the dose 1.0 mg l -1 of TDZ in the end of 90th day. The best result in terms of rooting (14.8 nr) was obtained from the dose 0.4 mg l -1 of NAA from hypocotyl. The highest vigor rate (96.9 %) was obtained from the dose 1.5 mg l -1 of TDZ in the end of 90th day.

The health and vitality of callus growth is one of the prerequisites for the success of further in vitro studies. This study investigated the efficiency of different percentage (0%, 10%, 20%, 30%, and 40%) of vermicompost tea as an organic substance on in vitro callus growth in sainfoin. Morpho-physiological responses of calli to vermicompost tea measured under in vitro conditions. As a result of this investigation, a combination of plant growth regulators (4 mg/l BAP and 0.5 mg/l NAA) with 20% of vermicompost tea causing significant callus initiation and growth in sainfoin stem explants. Under the light of these scientific findings, vermicompost tea might be used as an organic bio stimulant for efficient callus growth and complementary to commercial chemical hormones in sainfoin. This research is important due to it can contribute positively to the plant species that are difficult in terms of callus growth and plant regeneration in tissue culture.
