Guidance to rational use of pharmaceuticals in gallbladder sarcomatoid carcinoma using patient-derived cancer cells and whole exome sequencing

Abstract: PurposeGallbladder sarcomatoid carcinoma is a rare cancer with no clinical standard treatment. With the rapid development of next generation sequencing, it has been able to provide reasonable treatment options for patients based on genetic variations. However, most cancer drugs are not approval for gallbladder sarcomatoid carcinoma indications. The correlation between drug response and a genetic variation needs to be further elucidated.Experimental DesignThree patient-derived cells-JXQ-3D-001, JXQ-3D-002, and … Show more

“…A number of drugs for screening were limited in the actionable mutations and related pathways 2528 . In this study, only targeted deep sequencing focused on tumor-related genes was an effective approach to identifying genomic variants associated with cancer tumorigenesis in a large cohort of ESCC and their PDCs.…”

“…Under the microscope, the tumor tissue contained two kinds of epithelioid components and sarcomatoid spindle cell components, or no epithelioid components, and the morphology was similar to spindle cell sarcoma but actually was carcinoma [6,7]. Immunohistochemical examination showed positive markers of epithelial origin, like CK and EMA, and mesenchymal origin, like Vim and SMA, in spindle cell components [8][9][10]. In this case, upper abdominal pain, nausea and vomiting were the main manifestations of admission, and upper abdominal tenderness and Murphy's positive signs were present.…”
