GUILDify v2.0: A Tool to Identify Molecular Networks Underlying Human Diseases, Their Comorbidities and Their Druggable Targets

Abstract: The genetic basis of complex diseases involves alterations on multiple genes. Unravelling the interplay between these genetic factors is key to the discovery of new biomarkers and treatments. In 2014, we introduced GUILDify, a web server that searches for genes associated to diseases, finds novel disease-genes applying various network-based prioritisation algorithms and proposes candidate drugs. Here, we present GUILDify v2.0, a major update and improvement of the original method, where we have included protei… Show more

“…To corroborate these biomarkers, we tested how many of them are found using a different approach also based on the use of functional networks. For this purpose, we used GUILDify v2.0 [23], a web server that extends the information of disease-gene associations through the protein-protein interactions network. GUILDify scores proteins according to their proximity with the genes associated with a disease (seeds).…”

“…We then compared these sets of MoAs and proposed a list of biomarkers to identify potential cases of MD when using sacubitril/valsartan. Simultaneously, we used GUILDify v2.0 web server [23] as an alternative approach to compare the biomarkers proposed by TPMS and reinforce the results.…”

“…There are already several examples of repurposed drugs identified using TPMS approach that have been validated in vitro and in vivo [ 8 , 9 , 12 18 ] with one of them advancing to clinical trials. Simultaneously, GUILDify v2.0 web server [ 19 ] has been used as an alternative method to corroborate the promising combinations.…”

“…In the following sections, we analysed and compared the four groups of MoAs and searched for biomarkers that can potentially identify MD as an adverse effect of the drug (and consequently classify patients). We used the web server GUILDify v2.0 15 to analyse the comorbidity of both phenotypes and analyse the relevance of the proposed biomarkers in a different context.…”
