We present an up-to-date analysis for a precise determination of the effective fine structure constant and discuss the prospects for future improvements. We advocate to use a determination monitored by the Adler function which allows us to exploit perturbative QCD in an optimal well controlled way. Together with a long term program of hadronic cross section measurements at energies up to a few GeV, a determination of α(MZ ) at a precision comparable to the one of the Z mass MZ should be feasible. Presently α(E) at E > 1 GeV is the least precisely known of the fundamental parameters of the SM. Since, in spite of substantial progress due to new BaBar exclusive data, the region 1.4 to 2.4 GeV remains the most problematic one a major step in the reduction of the uncertainties are expected from VEPP-2000 [1] and from a possible "high-energy" option DAFNE-2 at Frascati [2]. We propose to measure the threshold lepton asymmetry, that is the forward-backward asymmetry of the charged lepton in tt events near the production threshold. At threshold top quark pairs are produced in an s-wave. Angular momentum conservation then implies that the top spins equal the spin of the initial state which -in the case of quarks -is uniquely fixed by the chirality of the initial quarks. Thus measuring final state top spins determines the chirality of the quarks which produced them. Information about the top spins can be extracted by measuring the angular distribution of the charged lepton in semileptonic or dileptonic decays of the top pair. One such distribution, the threshold lepton asymmetry, vanishes in tree-level QCD but is non-zero if new physics modifies the relative contribution of right-handed and left-handed quarks to top pair production. This is interesting because realistic models addressing the anomalous tt asymmetry have chiral couplings to light quarks. Models with identical tt asymmetries at the Tevatron can be distinguished by their threshold lepton asymmetries which range between plus and minus 25% in realistic models.

Salmonella typhimurium employs the specialized type III secretion system encoded in pathogenicity island 1 (SPI1) to translocate effector proteins into host cells and to modulate host cell signal transduction. The SPI1 type III system and the effector proteins are conserved among all salmonellae and are thought to be acquired by horizontal gene transfer. The genetic mechanisms mediating this horizontal transfer are unknown. Here, we describe that SopE, a SPI1-dependent translocated effector protein, is present in relatively few S. typhimurium isolates. We have isolated a temperate phage that encodes SopE. Phage morphology and DNA hybridization, as well as partial sequence information, suggest that this phage (SopE⌽) is a new member of the P2 family of bacteriophages. By lysogenic conversion this phage can horizontally transfer genes between different S. typhimurium strains. Strikingly, most of the isolates harboring SopE⌽ belong to the small group of epidemic strains of S. typhimurium that have been responsible for a large percentage of human and animal salmonellosis and have persisted for a long period of time. Our data suggest that horizontal transfer of type III dependent effector proteins by lysogenic infection with bacteriophages (lysogenic conversion) may provide an efficient mechanism for fine-tuning the interaction of Salmonella spp. with their hosts.
