HA/HSA co-modified erlotinib–albumin nanoparticles for lung cancer treatment

Abstract: BackgroundAim of this study was to prepare the hyaluronic acid and human serum albumin modified erlotinib nanoparticles (ERT-HSA-HA NPs) delivery system by a precipitation method.MethodsERT-HSA-HA NPs were characterized for physical properties, such as morphology and particle size, and in vitro drug release. Moreover, the cytotoxicity, cellular uptake, in vivo studies of ERT-HSA-HA nanoparticle were investigated and compared in A549 cells.ResultsThe ERT-HSA-HA NPs showed spherical morphology, and their hydrody… Show more

“…Lipid-based nanoparticles have been used to deliver erlotinib [ 48 ] and afatinib [ 49 ] and are shown to have enhanced therapeutic efficacy against NSCLC. Similarly, hyaluronic acid-based nanoparticles have been explored for the delivery of erlotinib [ 50 ] and afatinib [ 51 ] to target tumors.…”

“…This protein improves the release of drug bioavailability and decreases drug resistance in the body [ 29 ]. It is non-toxic and non-immunogenic [ 30 ]. Albumin-bound technology (named nab-technology) was developed and used for the delivery of drugs in the treatment of various types of tumors [ 31 ].…”

“…The pharmacokinetic parameters (blood residence time) in vivo of HA-decorated NPs were similar to that of ERT-HSA NPs. However, in terms of in vivo antitumor activity, mice treated with ERT-HSA-HA NPs showed a significantly suppressed tumor growth and no relapse after 30 days of treatment [ 173 ].…”
