Schizophrenia is a chronic, severe and disabling psychiatric disorder, whose treatment is based on psychosocial interventions and the use of antipsychotic drugs. While the effects of these drugs are well elucidated in neuronal cells, they are still not so clear in oligodendrocytes, which play a vital role in schizophrenia. Thus, we aimed to characterize biochemical profiles by proteomic analyses of human oligodendrocytes (MO3.13) which were matured using a protocol we developed and treated with either haloperidol (a typical antipsychotic), clozapine (an atypical antipsychotic) or a clozapine + d-serine co-treatment, which has emerged lately as an alternative type of treatment. this was accomplished by employing shotgun proteomics, using nanoeSi-Lc-MS/MS labelfree quantitation. Proteomic analysis revealed biochemical pathways commonly affected by all tested antipsychotics were mainly associated to ubiquitination, proteasome degradation, lipid metabolism and DNA damage repair. Clozapine and haloperidol treatments also affected proteins involved with the actin cytoskeleton and with EIF2 signaling. In turn, metabolic processes, especially the metabolism of nitrogenous compounds, were a predominant target of modulation of clozapine + d-serine treatment. in this context, we seek to contribute to the understanding of the biochemical and molecular mechanisms involved in the action of antipsychotics on oligodendrocytes, along with their possible implications in schizophrenia. BackgroundMedication discontinuation studies explore the outcomes of stopping a medication compared to continuing it. Comprehensively identifying medication discontinuation articles in bibliographic databases remains challenging due to variability in terminology.ObjectivesTo develop and validate search filters to retrieve medication discontinuation articles in Medline and Embase.MethodsWe identified medication discontinuation articles in a convenience sample of systematic reviews. We used primary articles to create two reference sets for Medline and Embase, respectively. The reference sets were equally divided by randomization in development sets and validation sets. Terms relevant for discontinuation were identified by term frequency analysis in development sets and combined to develop two search filters that maximized relative recalls. The filters were validated against validation sets. Relative recalls were calculated with their 95% confidences intervals (95% CI).ResultsWe included 316 articles for Medline and 407 articles for Embase, from 15 systematic reviews. The Medline optimized search filter combined 7 terms. The Embase optimized search filter combined 8 terms. The relative recalls were respectively 92% (95% CI: 87–96) and 91% (95% CI: 86–94).ConclusionsWe developed two search filters for retrieving medication discontinuation articles in Medline and Embase. Further research is needed to estimate precision and specificity of the filters.

Schizophrenia, a fare mental disorder that affects up to 1% of the population among the world, is a significant topic being researched in recent years. Some researchers have found a gene called Neuregulin-1(NRG1) that may relate to schizophrenia. However, the role of NRG1 in schizophrenia has not yet been well explained. Therefore, this research essay collects information on NRG1 and schizophrenia and then analyzes the effects of NRG1 on neurons, such as excitatory/inhibitory neurotransmission. This essay would also focus on the treatments of schizophrenia, including pharmacological therapy and CRISPR-based evaluation of schizophrenia. In this case, the essay would research on the relationship between NRG1 and schizophrenia. As research shows, the NRG1 may negatively lead to schizophrenia genetically. The interaction between NRG1 and other substances in our brain, such as NMDAR or GABA released by interneurons. Treatments for schizophrenia could deal with this disease through two main pathways; nonpharmacological therapy and pharmacological therapy could help with easing the patient by releasing the symptoms. CRISPR, however, could deal with schizophrenia directly by editing a certain section of DNA. So people could either release the symptoms causedthe by the interaction of NRG1 with other receptors, or edit DNA information by CRISPR.
