Harnessing the Benefits of Endogenous Hydrogen Sulfide to Reduce Cardiovascular Disease

Abstract: Cardiovascular disease is the leading cause of death in the U.S. While various studies have shown the beneficial impact of exogenous hydrogen sulfide (H2S)-releasing drugs, few have demonstrated the influence of endogenous H2S production. Modulating the predominant enzymatic sources of H2S—cystathionine-β-synthase, cystathionine-γ-lyase, and 3-mercaptopyruvate sulfurtransferase—is an emerging and promising research area. This review frames the discussion of harnessing endogenous H2S within the context of a non… Show more

“…From a clinical perspective, H 2 S has been posited to have a protective role against the onset and development of atherosclerosis 62 64 . Whereas defects in H 2 S signalling, including its synthesizing enzymes, can lead to CVD and associated complications 15 , 65 68 , H 2 S-based interventions have proved to be beneficial in preventing adult-onset CVD in animal studies via the reversal of disease-programming processes 69 . Plasma H 2 S levels have been shown to be significantly lower in patients with coronary heart disease than in angiographically normal control individuals 70 .…”

“…H 2 S concentration is an important determinant of the body's defense against disease. Dysregulation of H 2 S levels has been described in the pathogenesis of various conditions ranging from cardiovascular diseases to septic shock [ 316 , 317 ]. Reduced H 2 S levels are proposed as a contributor to the pathogenesis of asthma, and rescue with exogenous H 2 S was found to alleviate asthma severity in two separate animal models [ 318 , 319 ].…”

“…Increasing knowledge of the biochemistry of H 2 S may be exploited for the design of dietary intervention studies aimed at augmenting antioxidant defense. Detailed reviews on the roles of H 2 S in antioxidant functions with potential clinical relevance are available [150,[152][153][154]. A review of chemical tools to study small molecule persulfides has also been published [155].…”
