Heme oxygenase-1 accelerates erastin-induced ferroptotic cell death

Abstract: The oncogenic RAS-selective lethal small molecule Erastin triggers a unique iron-dependent form of nonapoptotic cell death termed ferroptosis. Ferroptosis is dependent upon the production of intracellular iron-dependent reactive oxygen species (ROS), but not other metals. However, key regulators remain unknown. The heme oxygenase (HO) is a major intracellular source of iron. In this study, the role of heme oxygenase in Erastin-triggered ferroptotic cancer cell death has been investigated. Zinc protoporphyrin I… Show more

“…A candidate family of enzymes with this property is the family of lipoxygenases that catalyze the dioxygenation of polyunsaturated fatty acids in lipids [86]. In addition, the iron-containing heme oxygenase-1 (HO-1) was reported to be induced upon erastin treatment and to accelerate erastin-induced ferroptosis[87]; thus, HO-1 may be a source of iron driving the lipid peroxidation that occurs during ferroptosis, at least in response to erastin.…”

“…HMOX1 is necessary for ferroptosis and is a major source of iron in the body. [29] Inhibition of cysteine-glutamate exchange through System x c- , of which SLC7A11 is a component, induces iron-dependent cell death. [30] To determine whether 35G8 induces ferroptosis in U87MG cells, we treated the cells in the presence or absence of deferoxamine (DFO), an iron chelator (Figure S2B).…”

“…[29, 30] SLC7A11 is a negative regulator of ferroptosis and upregulation of SLC7A11 occurs as a response to system x c- inhibition. [13] Efforts to treat glioma patients by inhibiting system x c- have failed; [45] however, combining SLC7A11 inhibition with a PDI inhibitor may be a promising new strategy.…”
