Hemodynamic Assessment of the Development of Portal-Systemic Collaterals in Portal Hypertensive Rats

Abstract: We concluded that the pressure-induced opening of pre-existing vessels was not the primary underlying mechanism in the formation of SRS. Endothelial proliferating and vascular remodeling process participated actively during the development of SRS. These observations can be used for studying the pathogenesis and developing more effective anti-portal hypertensive therapy in the future.

“…PHT induces the formation of portal systemic collaterals. Revealing the developmental change in portal-systemic collaterals is important for future therapy [ 9 ]. The incidence of gastric varices is 25 % and the rate of gastric variceal hemorrhage is less than esophageal variceal hemorrhage [ 10 ].…”

“…The resin was allowed to polymerize intrahepatically for 72h, before being macerated for about 5 days in potassium hydroxide (25% KOH). The vascular replicas were µCT imaged at a resolution of [32][33][34][35][36][37][38][39][40] µm using a high-resolution µCT scanner (HECTOR, Centre for X-ray Tomography (UGCT), Ghent University, Belgium) to acquire detailed 3D datasets of the hepatic vasculature. A more elaborate description of the VCC and µCT protocol was described earlier [18].…”

“…An extrahepatic shunt was added to the model by means of a vascular resistance and guides part of the blood from the digestive organs directly into the systemic venous system, thereby bypassing the liver [15,40,41]. During the morphological analysis, two portosystemic shunts were detected in one of the cirrhotic rats (cirrhosis 2) (illustrated in [22]), shunting blood directly from the portal trunk into the caudal vena cava.…”

“…Данные патофизиологические нарушения могут стать начальным звеном в развитии портосистемного коллате-рального кровообращения при ПГ [25]. На поверхности активированных эндотелиальных клеток происходит ад-гезия моноцитов, которые продуцируют факторы роста и протеиназы, такие как урокиназный активатор плазми-ногена и матриксные металлопротеиназы, способствуя ми-грации и делению гладкомышечных клеток.…”
