Hepatoprotective Effect of Citrus aurantium L. Against APAP-induced Liver Injury by Regulating Liver Lipid Metabolism and Apoptosis

Abstract: Acetaminophen (APAP) refers to a medication used to manage pain and fever symptoms, but it always causes liver injury when overdosed. Zhishi, dried young fruit of Citrus aurantium L., is a famous Citrus herbal medicine in Asian countries which is rich in dietary phenolic substances. In this study, the mechanism of Zhishi protected against APAP-induced liver injury was studied more deeply by metabolomic strategy and pharmacological study. The metabolomics results demonstrated that Zhishi can prevent the APAP-in… Show more

“…The APAP-induced upregulation of Bax is attenuated by the inhibition of JNK activation [19,23,24]. APAP has been reported to induce and inhibit the expression of Bax (pro-apoptotic factor) and Bcl-2 (anti-apoptotic factor), respectively, and the upregulation of the Bax/Bcl-2 ratio damages the mitochondrial membrane and induces a caspasedependent apoptosis pathway [24,25], with resulting hepatocyte death that could promote the onset and progression of liver disease [25]. The findings of the present study indicate that APAP overdose induces typical signs of apoptosis, including caspase 1 activation and DNA fragmentation.…”

“…Recently, herbs with homology of medicine and food have been further investigated worldwide [5], Mulberry leaves (dried leaves of Morus alba L . [Moraceae family]) are approved by the Ministry of Health of the People’s Republic of China as “medicine food homology” materials in 1991 [6].…”

“…Regarding the peels, flavonols and anthocyanins with antioxidant activity have been described [ 17 ]. There are several animal models to evaluate the hepatoprotective effect of natural products; one of the effects is the model of liver damage induced by acetaminophen (APAP), a drug widely used as an antipyretic and analgesic, which in high doses can produce necrosis and insufficiency acute hepatica [ 19 , 20 , 21 ].…”
