Heptapeptide Isolated from Isochrysis zhanjiangensis Exhibited Anti-Photoaging Potential via MAPK/AP-1/MMP Pathway and Anti-Apoptosis in UVB-Irradiated HaCaT Cells

Abstract: Marine microalgae can be used as sustainable protein sources in many fields with positive effects on human and animal health. DAPTMGY is a heptapeptide isolated from Isochrysis zhanjiangensis which is a microalga. In this study, we evaluated its anti-photoaging properties and mechanism of action in human immortalized keratinocytes cells (HaCaT). The results showed that DAPTMGY scavenged reactive oxygen species (ROS) and increase the level of endogenous antioxidants. In addition, through the exploration of its … Show more

“…Peptide fraction from Pinctada martensii meat containing oligopeptides FH, AL, MY, AGF, and IYP showed the anti-photoaging activity by increase cell viability, reduced the interstitial MMP-1 and MMP-3 contents, and downregulated the expression of p38, EKR, JNK, MMP-1, and MMP-3 in UVB-induced HaCaT cells [ 58 ]. Heptapeptide DAPTMGY from Isochrysis zhanjiangensis showed protective effects on HaCaT cells against UVB-induced damage through regulating anti-apoptosis and MAPK/AP-1/MMP pathway [ 59 ]. Hydrolysate (TCH) from Theragra chalcogramma was rich in GLPYT and could alleviates photoaging via controlling the deposition of collagen fibers and recovery of extracellular component matrix in SD rats [ 60 ].…”

“…In this study, we investigated several gene markers that we selected through a whole-transcriptome search for differential expressions. We found differentially expressed genes related to apoptosis and immune responses to inflammation, such as FOXO3, TLR4, caspase-8, MMP-9, and p38 MAP kinase (p38) ( Cuadrado and Nebreda, 2010 ; van der Vos and Coffer, 2011 ; Zheng et al, 2021 ). In addition, we investigated the effects of DDK-401 and DDK-204 without any stimulation (UV-B irradiation) in HaCaT cells to observe whether our samples could trigger inappropriate apoptosis or inflammation under normal conditions showed in ( Figure 7 ).…”

“…The results of the current study indicated that hUC-MSC-CM inhibited UVB-induced HaCaT cell apoptosis, ROS production, and promoted cell proliferation and migration. Apoptosis is affected by multiple signalling pathways, including the p53 signalling pathway [56]. p53 was detected in both transcriptomics data and western blot experiments, revealing that p53 expression was increased after UVB irradiation and decreased after the addition of hUC-MSC-CM.…”
