Human T cell clones specific for tetanus toxoid (TT) or for alloantigens were isolated and expanded in culture using antigen stimulation and soluble growth factors. When stimulated by the specific antigen, the clones were able to proliferate in the absence of exogenous growth factors and to provide help to B cells. The alloreactive and TT-specific clones were compared for their capacity to help an anti-TT as well as a polyclonal antibody response. The frequency of the B cells activated to the production of specific antibody was determined by limiting dilution analysis in cultures containing limiting numbers of responding B cells and optimal numbers of cloned T helper cells. In these conditions a single activated b cell was able to produce about 10 ng of antibody. TT-specific clones, in the presence of low TT concentrations, selectively induced a number of TT-specific B cells to produce IgG antibody in the absence of a detectable polyclonal B cell activation. On the other hand the alloreactive clones activated higher numbers of TT-specific B cells as a part of a strong polyspecific B cell activation. In this case the antibody production did not require the presence of TT in culture; furthermore cells producing antibodies of unrelated specificities were also activated in allostimulated cultures. IgG anti-TT were produced only by immune donors, while IgM anti-TT were produced by immune and nonimmune donors, providing the B cells were activated by alloreactive clones. These data demonstrate that human memory B cells can be triggered by both antigen-specific and alloreactive T cells to antibody production. The alloreactive clones can therefore be used to analyze at the clonal level the repertoire of a pool of B cells that contain most, if not all, the memory antibody specificities.

Radioimmunoprecipitation followed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis was used to study the distribution on human lymphoid cells of a previously undescribed surface antigen recognized by several heteroantisera. A glycoprotein with a 90,000 mol wt (under reducing conditions) was detected on all cell lines tested including T, B, null, and myeloid cell lines, although the amount of antigen present varied considerably. The antigen was absent from normal peripheral blood lymphocytes (PBL), B and T cells, monocytes, granulocytes, thymocytes, and erythrocytes. After stimulation with lectins or allogeneic B cells, the antigen was induced on PBL T cells. A limited number of leukemic T cells tested all expressed the antigen, as did melanoma cell line and human embryonic lung fibroblasts. Hence, the antigen was present only on dividing lymphoid cells and absent from nondividing cells, but was also present on the two examples of dividing non-lymphoid cells tested. Under nonreducing conditions, the 90,000-mol wt band normally present disappeared to be replaced by another at approximately 200,000 mol wt. The glycoprotein bound to lectins from lentil and ricin, but not to wheat germ agglutinin. It could be readily labeled metabolically by [35S]methionine or by surface iodination, and appeared to be a major membrane protein on some cell lines.

A monoclonal antibody, PVR-1, was obtained after hybridization of X63Ag8.653 murine myeloma cells with spleen cells from a mouse immunized with human lymphocytes. It recognizes a 175,000-to 185,000-dalton surface antigen present on -"80% ofnormal human peripheral T lymphocytes, 50% ofnon-T non-B cells, and < 10% of B cells as determined by complementdependent microcytotoxicity. It is also present on various leukemia T cells, on some but not all T lymphoblastoid cell lines, and on a small fraction of some B lymphoblastoid cell lines. Some B-cell chronic lymphocytic leukemia cells also express the PVR-11 antigen. Functional analysis of normal human T lymphocytes demonstrated that the PVR-11-depleted T-cell subset contains the precursors ofboth cytotoxic and suppressor cells but lacks helper cells. On the other hand, cytotoxic effector T cells express the antigen. These results demonstrate that antigenic determinants with relatively wide tissue distribution can dissect functionally distinct human immunoregulatory T-cell subsets.During the last several years, a number of antibodies that recognize distinct surface membrane antigens on human T lymphocytes have been described.
