Heterogeneity of mitochondrial DNA haplotypes in Pre-Columbian natives of the Amazon region

Abstract: We report the first study of mitochondrial DNA (mtDNA) sequencing from ancestral Amerindian populations of the South American continent. Sequencing of the D-loop region of mtDNA was carried out for bone fragments from 18 skeletons of Pre-Columbian Amerinidians. The skeletons were excavated in different archeological sites of the Brazilian Amazon region, with dating estimated at 500-4,000 years before the present. The sequencing of at least 354 bases permitted the identification of 13 haplotypes defined by vari… Show more

“…An example of this loss is the X mitochondrial haplogroup. Nowadays it is completely absent among the Indians of South America (41), whereas a couple of millennia ago it could still be found in the subcontinent (42). More light on this subject will be certainly shed when Paleoindian DNA fragments are recovered and analyzed.…”

“…Among Amerindian populations, five main haplogroups are prevalent (A, B, C, D, and X- [21,22]) [13,[23][24][25][26][27]. Asian populations share American haplogroups (A, B, C, and D), as well as other haplogroups (M, N, G, F, Z, Y, and R) that are described as specific to the Asian continent [28,29].…”

“…The extraction and treatment of the DNA samples were performed in the Paleogenetic section of the Human and Medical Genetics Laboratory at the Federal University of Pará (UFPA) according to the established protocol [25,32]. The DNA was extracted either with the phenol-chloroform method [33] with modifications or with the DNA IQ System kit (Promega, Madison, WI, USA).…”

“…La presencia de secuencias no pertenecientes específicamente a algún haplogrupo (que comúnmente se clasifican como E) es un fenómeno frecuente en este tipo de estudios (13)(14)(15)18,19,23).…”
