“…Previous work has established the feasibility of both subcutaneous and intracranial sites of tumour transplantation in immune-deficient rodents (Rana et al, 1977;Bradley et al, 1978;Shapiro et al, 1979). The reason for using two different implantation sites was to evaluate the possibility that the intracranial microenvironment was unique with respect to oxygenation owing to differences in tumour interstitial pressure, tumour blood flow and/or vascular architecture.…”

“…The desire to experiment on human glioblastomas within a convenient in vivo system eventually led to the use of immunodeficient animals, most commonly in athymic nude mice or rats, which can serve as tolerant hosts for human neoplasms (Rana et al 1977). The resulting tumors are interspecific transplantation chimeras and therefore constitute experimental systems in which the relationship between the tumor cells and the host tissues can be easily analyzed.…”

“…In most previously published orthotopic animal models of human GBM (Rana et al 1977;Bradley et al 1978;Shapiro et al 1979;Bullard et al 1981;Horten et al 1981;Jones et al 1981;Basler and Shapiro 1982;Marno et al 1982;De Armond et al 1994), the tumors were implanted not in solid pieces but by stereotaxic injection of cell suspensions. The cell suspension methods of grafting have the distinctive advantage of limited surgical trauma.…”

“…Although the growth of 2 human glioblastomas and 1 mengingioma in mice has been described previously from these laboratories and elsewhere (Cobb and Mitchley, 1974;Rana et at., 1977) the present study deals with a larger series of the type of gliomas that can be expected to grow. Our preliminary observations suggest that such tumours in immune-reduced mice respond to chemotherapeutic agents (CCNU, BCNU) which have been successful in man.…”

“…We hope that the methods derived will complement studies on murine ependymomas (Ausman et al, 1970) and rat astrocytomas (Benda et al, 1971;Rosenblum et al, 1975 (Cobb and Mitchley, 1974) based on an earlier method of Miller (1963). Briefly, mice were thymectomised 4-6 weeks before exposure to a potentially lethal dose of whole body irradiation (900 rad from a 220 kV X-ray machine).…”
