Heterotypic signaling of cancer-associated fibroblasts in shaping the cancer cell drug resistance

Abstract: The context-dependent reciprocal interaction between the cancer cells and surrounding fibroblasts is imperative for regulating malignant potential, metabolic reprogramming, immunosuppression, and ECM deposition. However, recent evidence also suggests that cancer-associated fibroblasts induce chemoresistance in cancer cells to various anticancer regimens. Because of the protumorigenic function of cancer-associated fibroblasts, these stromal cell types have emerged as fascinating therapeutic targets for cancer. … Show more

“…The reciprocal interaction between tumor cells and fibroblasts plays a crucial role in the progression of breast cancer [104,105]. OPN emerges as a key regulator in facilitating the crosstalk between tumor and stromal fibroblasts, contributing to breast cancer advancement [62,104,106].…”

“…CAFs are represented by highly heterogeneous cell populations which account for about 50% of the total number of all TME cells ( Butti et al, 2023 ; Wieder, 2023 ). Therefore, CAFs are often defined as stromal TME cells ( De et al, 2021 ; Kochetkova and Samuel, 2021 ; Mor et al, 2022 ).…”

“…The TME components consist of an extracellular matrix (ECM) and multiple cell types including cancer‐associated fibroblasts (CAFs), tumor‐associated macrophages (TAMs), endothelial cells, and mesenchymal stem cells (MSCs) 256 . Tumor growth and progression and subsequent development of chemoresistance are mediated by the cross‐talk between tumor cells and their surrounding stromal cells via paracrine loops, chemokine networks, and cell–cell interactions 257–260 . CAFs are one of the most important nonmalignant cell populations in the TME, and they play a crucial role in cancer progression, metastasis and chemotherapy resistance 261,262 .…”
