HIF-1α inhibition by 2-methoxyestradiol induces cell death via activation of the mitochondrial apoptotic pathway in acute myeloid leukemia

Abstract: The bone marrow microenvironment plays an important role in the development and progression of AML. Leukemia stem cells are in a hypoxic condition, which induces the expression of HIF-1α. Aberrant activation of HIF-1α is implicated in the poor prognosis of patients with acute myeloid leukemia (AML). Herein, we investigated the expression of HIF-1α in AML and tested 2-methoxyestradiol (2ME2) as a candidate HIF-1α inhibitor for the treatment of AML. We found that HIF-1α was overexpressed in AML. HIF-1α suppressi… Show more

“…A synergy was observed between metformin, which inhibits Complex I in mitochondria, and the broad‐spectrum metabolic inhibitor 6‐benzylthioinosine (6‐BT), with reduced glycolysis, ROS suppression and increased apoptosis (Sabnis et al , ). Inhibition of HIF1α by 2‐methoxyestradiol was found to activate the mitochondrial apoptotic pathway in AML (Zhe et al , ). Inhibitors for histone deacetylases as well as protein deacetylase SIRT1 have been evaluated in clinical trials in the US and European Union.…”

“…The latest reports about the mechanism of action of 2-ME2 offer further insight into the importance of its influence on HIF-1α function, supported by the observations that when 2-ME2 is combined with other conventional chemotherapeutics or radiation therapy, it can increase the anticancer activity of the concomitant agent(s) or that of the irradiation [35][36][37]. Moreover, based on its inhibitory effect on HIF-1α acute myeloid leukemia has also been identified as a possible novel therapeutic target for 2-ME2 [38].…”

“…2-ME-mediated mitochondrial apoptotic pathway activation is mediated by reactive oxygen species (ROS) in acute myeloid leukemia cells. 37 Furthermore, Foster et al 38 indicated that 1 µM 2-MeE2bi-sMATE caused loss of mitochondrial membrane potential followed by increased caspase-3 and caspase-7 activation in ovarian carcinoma (A2780), LNCaP and MCF-7 cells.…”
