High Concentrations of Soluble Tumor Necrosis Factor Receptors in Ascites

Abstract: Ascites and plasma concentrations of soluble tumor necrosis factor receptors p55 and p75 were measured in a prospective study in 34 patients (35 occasions of ascites) with hepatic (5 infected and 21 uninfected) and malignancy-related (9) ascites. All patients had high concentrations of both soluble tumor necrosis factor receptors in ascites and plasma; these were about 500 times higher than the corresponding tumor necrosis factor-alpha concentrations. Ascites levels of soluble tumor necrosis factor receptors p… Show more

“…Interleukin-6 has been found to be produced in high amounts in the peritoneal cavity of patients with hepatic or malignant ascites. 22 High concentrations of soluble TNF receptors have been found in ascites 22 and may explain why, in recent studies, malignant ascites responded to bolus intraperitoneal injections of TNF. 23 Administration of interferon-alpha 2b also has been shown to reduce ascites.…”

“…It has been reported that physiologic inhibitors for TNF are present in a variety of body fluids including serum [1,24], urine [23,24], ascites [2], and also in the epithelial lining fluids of the respiratory tract [31]. These inhibitors are soluble TNF receptors (sTNF-Rs) that were cleaved from the extracellular domain of membrane sTNF-Rs.…”

“…Minn., USA). The sTNFRs were determined with an enzyme-linked immunological and biologi cal binding assay (EL1BA) as reported by Andus et al [25], Briefly, samples or sTNFRs standards and recombinant human TNF-a labeled with peroxidase were added to the well of a microtiter plate precoated with non-neutralising monoclonal (mouse) antibodies to sTNFRs (clone htr 20 for the sTNFR-B and clone utr 4 for the sTNFR-A). After incubation for 16-24 h at I5-25°C and a washing step, the peroxidase bound to the microtiter plate was measured enzymatically.…”
