Summary Híbrido de Timor (HT) is a natural interspecific hybrid of Coffea arabica L. with Coffea canephoraPierre ex Froehner that has played a substantial role in breeding programs as source of resistance genes. The original HT plant is represented by the anorthoploid accession CIFC 4106 , an allotriploid plant with 3x=33 chromosomes and 2C=2.10 pg. From this HT plant, other accessions have been obtained and used in crossings with C. arabica to provide resistant plants to the main pathogens. However, each HT accession and their derivates show a particular karyotype due to irregular anaphasic disjunction, which results in a few reproductive cells with an unbalanced number of chromosomes. Regarding this fact and the crop relevance of HT, this study aimed to develop a direct somatic embryogenesis system for the clonal propagation of this germplasm, using the first HT plant as explant donor. To accomplish this purpose, disinfested leaf explants of greenhouse-cultivated HT CIFC 4106 plants were inoculated in medium supplied with 0.001 g L 1 6-benzylaminopurine. Somatic embryogenesis process was asynchronous, with distinct developmental stages occurring simultaneously. Besides, emergence of secondary embryos from primary ones was observed. Mature embryos were germinated, and well-rooted embryos were selected for plantlet regeneration. The nuclear DNA content and the karyotype showed that allotriploidy was conserved in all regenerated plantlets. Considering these results, the direct somatic embryogenesis protocol adopted in the present work was imperative for the accurate clonal propagation of HT, being relevant for multiplication and conservation of elite accessions that show an irregular meiosis.

“…De acordo com a Figura 5, maiores freqüências de CEF s foram obtidas com a utilização de 1 mg.L -1 de 2,4-D com concentrações crescentes de BAP, seguindo uma tendência linear, sugerindo, portanto, a possibilidade de incrementar concentrações de BAP acima de 8 mg.L -1 , com o objetivo de aumentar a freqüência de CEF s. Resultados semelhantes foram obtidos com a concentração de 8 mg.L -1 de BAP sem a adição ao meio de cultura de 2,4-D Segundo Sondahl & Sharp (1977), a ação combinada de auxina/citocinina é fundamental para obtenção de calos embriogênicos friáveis em Coffea, o que foi posteriormente comprovado por Garcia & Menendez (1987) em explantes foliares de Catimor. Os resultados obtidos no presente experimento foram semelhantes à-queles apresentados por Cordeiro (1999), que verificou um efeito negativo da auxina 2,4-D na freqüência de CEF s, sendo similares ao efeito inibidor de ANA em explantes foliares da variedade Typica, em meio contendo BAP (Yasuda, et al, 1995).…”

INDIRECT SOMATIC EMBRYOGENESIS INCoffea arabica L. CV. OBATÃ 1 ABSTRACT It was aimed with this work to study the indirect somatic embryogenesis in Coffea arabica L. cv. 'Obatã', including the steps of induction, differentiation and regeneration of plants. Leaf segments withdrawn from plants under field conditions were disinfected with 70% alcohol for 1 , 1% sodium hypochlorite for 15 and inoculated in 'CI' medium (callus induction) supplemented with 2,4 D (0, 1, 2 and 4 mg.L -1 ) and kinetin (0, 2, 4 and 8 mg.L -1 ). Later, the callus were transferred to the 'CD' medium (callus differentiation) to which was added 2,4-D (0, 1, 2 and 4 mg.L -1 ) and BAP (0, 2, 4 and 8 mg.L -1 ). During the regeneration step, friable embryogenic callus were inoculated in 'R' medium supplemented with BAP (0, 2, 4 and 6 mg.L -1 ) and sucrose (0, 15, 30, 45 and 60 g.L -1 ). The culture media utilized had their pH adjusted to 5.6 ± 1 before being autoclaved. The experiments were kept in growth room at 26 ± 1 o C. Over the steps of callus 1.
