High Level Expression and Purification of the Clinically Active Antimicrobial Peptide P-113 in Escherichia coli

Abstract: P-113, which was originally derived from the human saliva protein histatin 5, is a histidine-rich antimicrobial peptide with the sequence AKRHHGYKRKFH. P-113 is currently undergoing phase II clinical trial as a pharmaceutical agent to fight against fungal infections in HIV patients with oral candidiasis. Previously, we developed a new procedure for the high-yield expression and purification of hG31P, an analogue and antagonist of human CXCL8. Moreover, we have successfully removed lipopolysaccharide (LPS, endo… Show more

“…Small-molecule AMPs were often expressed as fusion proteins [22,27]. A disadvantage of fusion protein expression for AMPs is that fusion proteins need to be processed in vitro, by proteases or chemicals, to release AMPs.…”

“…To obtain 15 N-, 13 C-labeled peptide for three-dimensional NMR analysis, P-113 was expressed in Escherichia coli BL21 (DE3) cells grown in M9 minimal media containing 15 N-labeled ammonium chloride (1 g/L) and 13 C-labeled glucose (2 g/L). Peptides were purified according to our previous study [32]. All NMR samples were prepared in SD broth containing 10% D 2 O.…”

“…Several efforts have been made to identify fragments of histatin 5 with pharmaceutical application and have yielded promising results. An example in this regard is the 12-amino acid peptide P113, which was evaluated in phase I and phase II clinical studies as pharmaceutical agent to fight oral candidiasis ( Woong et al, 2008 ; Cheng et al, 2018 ; Browne et al, 2020 ).…”
