High Mutation Frequency at Ha-ras Exons 1-4 in Squamous Cell Carcinomas from PUVA-treated Psoriasis Patients¶

Abstract: Clinical follow-up studies have revealed that PUVAtreated patients are at increased risk of skin cancer, particularly squamous cell carcinoma (SCC). However, since psoralen and UVA (PUVA) is not only a potent mutagen and carcinogen but also an immunosuppressor, and since other (co)carcinogenic factors often exist in psoriasis patients, the exact causes and mechanisms of PUVA-associated SCC are still not completely understood. In order to fill this gap the tools of molecular epidemiology are being used to study… Show more

“…The most commonly used carcinogens in mouse tumor induction are UV irradiation, DB[a,l]P, BaP, DMBA, TPA, nitric oxide and=or a combination of them. 4,12,32,[38][39][40][41][42][43] Although mutations were also found in N-ras 40 and k-ras 17 genes, most occurred in H-ras. A barely detectable mutation, GfiA at codon 12, was induced by UVA (Fig.…”

“…11 H-ras mutations are present in a large proportion of PUVAassociated SCCs. 12 BaP is often used as an index of environmental PAH levels, and its level in the environment is usually below the carcinogenic threshold. Thus, the combined impact of these 2 environmental factors at subcarcinogenic doses remains a challenging subject.…”

“…Recent studies show that there are 18 Ha-ras missense or nonsense mutations in this gene locus. 5 Overall, SCC is uncommon in dark-skinned individuals, but it is the most common cutaneous cancer in African-Americans. In such individuals, the cancer usually occurs in areas that are not sun-exposed.…”

“…Patrick Antony Oberholzer presented provisional results from a genetic screen for mutations in cutaneous SCCs. Genetic lesions identified to date in human cutaneous SCC include an activating mutation at position 12 of H-RAS (35-46%) (8,9) but not K-RAS or N-RAS, mutations and deletions at the locus of the tumor suppressor CDKN2A (9-42%) (10,11), and mutations in the gene encoding the tumor suppressor p53 (TP53; 41-69%) (12)(13)(14). Screening for somatic mutations was performed in SCCs from 20 OTRs and 20 nontransplant recipients.…”
