High performance liquid chromatography evaluation of serum carbohydrate-deficient transferrin and more sialylated transferrin glycoforms in children

Abstract: Age-specific Disialo-Tf cut-offs are not necessary. In children 1.3% and 6.4% may be suggested as upper limits of normal range to detect increases of Disialo- and Trisialo-Tf. The presence of Asialo- and Monosialo-Tf should be considered an abnormal finding and prompt further investigations.

“…This is due to the fact that probably the glycoform with a lower content of sialic acid are more saturated with iron, useful for the synthesis of hemoglobin which carries oxygen to the foetus. The distribution of disialotransferrin and other glycoforms with the highest content of sialic acid has been also studied in children and adolescents [13].…”

“…Ciò è dovuto al fatto che probabilmente le forme a più basso contenuto di acido sialico sono quelle più sature di ferro utile alla sintesi dell'emoglobina che porta ossigeno al feto. E' stata studiata anche la distribuzione della disialotransferrina e delle altre glicoforme a più alto contenuto di acido sialico nei bambini e adolescenti (Bianchi et al 2012).…”

“…Variations in the number of terminal sialic acid residues influence the electrophoretic mobility of the Tf molecule and thus permit analytic separation of the different isoforms . Among the human Tf isoforms, which vary from asialo‐Tf and octasialo‐Tf, the most common distribution observed in nonalcohol drinkers is tetra‐Tf (79.9%), followed by pentasialo‐Tf (14.0%), trisialo‐Tf (4.02%), disialo‐Tf (1.14%), and hexasialo‐Tf (0.73%) in order . In contrast, chronic alcohol abusers show an increase in the disialo‐Tf and asialo‐Tf fractions, which constitute the so‐called carbohydrate‐deficient transferrin (CDT).…”
