Background: Rhomboid domain containing 1 (RHBDD1) plays a crucial role in tumorigenesis. Silibinin, which is a natural extract from milk thistle, has shown antitumor effects against various tumors. Here, we investigate whether silibinin affects the function of RHBDD1 in non-small cell lung cancer (NSCLC) cell proliferation, migration and invasion. Methods: The Oncomine database and an immunohistochemistry (IHC) assay were used to determine the RHBDD1 expression levels in lung cancer tissues. The associations between RHBDD1 and overall survival rate or clinicopathological parameters were respectively assessed using the Kaplan-Meier overall survival analysis or Chi-squared test. CCK-8 and Transwell assays were applied to analyze cell proliferation, migration and invasion. A549 cells were incubated with increasing concentrations of silibinin. RHBDD1 knockdown and overexpression were achieved via transfection with si-RHBDD1 or RHBDD1 overexpression plasmid, respectively. Western blotting was performed to measure the expressions of epithelialmesenchymal transition (EMT) markers. Results:We found that overexpression of RHBDD1 in lung cancer tissues correlates with a poor prognosis of survival. Clinical specimen analysis showed that upregulation of RHBDD1 correlates remarkably well with TNM stage and lymph node metastasis. Silibinin suppresses A549 cell proliferation, migration, invasion and EMT in a dose-dependent manner. Importantly, RHBDD1 was downregulated in silibinintreated A549 cells. RHBDD1 overexpression reversed the suppressive effects of silibinin on A549 cell proliferation, migration, invasion and EMT expression, while its knockdown enhanced them.Conclusions: These findings shown an anti-tumor impact of silibinin on NSCLC cells via repression of RHBDD1.

Background/aims: MiR-216b and forkhead box M1 (FOXM1) were demonstrated to exert their biological effects on the development and progression of tumors. This study aimed to investigate the expression and role of miR-216b and FOXM1 in tissues and cell lines of non-small cell lung cancer (NSCLC). Methods: The expressions of miR-216b and FOXM1 in NSCLC tissues and cells were detected by qRT-PCR and Western blot analysis. Cell proliferation was measured by CCK-8 assay. Cell migration and invasion were confirmed by Transwell assay. Finally, the bioinformatics and dual-luciferase reporter assay were conducted to validate the relationship of miR-216b and FOXM1. Results: Compared with normal tissues and cells, the expression of miR-216b was obviously decreased in NSCLC tissues and cells. However, the expressions of FOXM1 mRNA and protein were significantly increased, and negatively correlated with the expression of miR-216b. Multivariate Cox’s regression analysis suggested that miR-216b or FOXM1 expression was an independent prognostic factor for patients with NSCLC. MiR-216b overexpression remarkably repressed cell proliferation, migration, invasion, and epithelial–mesenchymal transition (EMT) of NSCLC cells. The bioinformatics and dual-luciferase reporter assay validated that the 3ʹ-untranslated region (3ʹ-UTR) of FOXM1 mRNA was indeed a direct target of FOXM1. In vitro, overexpression of FOXM1 partially eliminated inhibitory effects of miR-216b on cell proliferation, migration, and invasion, whereas inhibition of FOXM1 contributed to inhibitory effects mediated by miR-216b. Conclusion: MiR-216b inhibits cell proliferation, migration, invasion, and EMT by targeting the expression of FOXM1 in human NSCLC. These findings suggested a potential therapeutic role of miR-216b in patients of NSCLC.

Background The role of Vasohibin-1 (VASH1) expression in solid tumors remains controversial. We thus performed this meta-analysis to elucidate the associations between VASH1 expression and the prognosis of solid tumors. Methods We searched relevant literature in PubMed, Web of Science and EMBASE. The hazard ratio (HR) or odds ratio (OR) and 95% confidence intervals (CIs) were measured by fixed-effects or random-effects models. Publication bias was assessed using funnel plots and Egger's regression test. Results The results showed that VASH1 expression exhibited a significantly decreased overall survival (OS) time (HR = 1.85; 95% Cl = 1.27–2.69) and disease-free survival (DFS) (HR = 1.80; 95% Cl = 1.41–2.29) time. Meanwhile, VASH1 expression was found significantly associated with TNM stage (OR = 1.96; 95% CI = 1.57–2.46), tumor stage (OR = 2.35; 95% CI = 1.88–2.93), lymph node metastasis (OR = 2.02; 95% CI = 1.37–2.98), venous invasion (OR = 1.63; 95% CI = 1.00-2.65), tumor grade (OR = 1.74; 95% CI = 1.13–2.67) and microvessel density (MVD) (OR = 4.30; 95% CI = 2.31–8.03). However, no significant association was found between the VASH1 expression and distant metastasis (OR = 1.81; 95% Cl = 0.74–1.41). Conclusion This study demonstrated that VASH1 expression was relevant to more aggressive clinicopathological parameters and might predict inferior DFS and OS in solid tumor patients.
