High-pressure liquid-chromatographic method for determination of gentamicin in plasma.

Abstract: A rapid, specific method for measuring gentamicin in plasma by high-pressure liquid chromatography was developed. After deproteinization, gentamicin in the supernate was dansylated and extracted into ethyl acetate. The organic extract was chromatographed on a microparticulate reversed-phase column, which was eluted with aqueous acetonitrile. Use of the dansyl derivative enables fluorometry, for more sensitive quantitation. Various factors that could affect the assay sensitivity were investigated. With 0.2-ml p… Show more

“…Other HPLC techniques report sensitivities of 0.2 mg/L of SA in urine 7 and 0.5 mg/L of SA in plasma. 14 A liquid chromatographic method with UV detection has a sensitivity of 0.5 mg/L with a precision of 8.6 mg/L but takes 25 min and requires purification steps. 15 Capillary electrophoresis coupled to laser-induced fluorescence has also been described to detect SA, SU, and other metabolites in urine following sample filtration and dilution.…”

“…For instance, the detection of gentamicin was accomplished with fluorescence detection after post‐column derivatization with 1,2‐phthalic dicarboxaldehyde (OPA) 9, 10. Similarly, liquid chromatography (LC) with pre‐column derivatization by OPA11 and dansyl chloride12 followed by fluorescence detection was performed for gentamicin analysis. Freeman et al 13.…”
