High-Resolution Native Mass Spectrometry

Abstract: Native mass spectrometry (MS) involves the analysis and characterization of macromolecules, predominantly intact proteins and protein complexes, whereby as much as possible the native structural features of the analytes are retained. As such, native MS enables the study of secondary, tertiary, and even quaternary structure of proteins and other biomolecules. Native MS represents a relatively recent addition to the analytical toolbox of mass spectrometry and has over the past decade experienced immense growth, … Show more

“…In contrast to the solution-based measurements of MP, CD-MS measurements are performed on particles following their ionization and introduction into the gas phase. Over the years, a body of evidence has been accumulated demonstrating that protein complexes can be mass analyzed in the gas phase while generally retaining their native stoichiometry and other aspects of higher-order structure . The similar results here obtained by MP and native CD-MS support the validity of the presented findings and the absence of measurement biases.…”

“…Furthermore, in MS analysis, mass accuracy and resolution typically diminish with increasing molecular weight, even when using the latest high-resolution mass spectrometers. 30 , 71 , 72 In addition, full sequence coverage is generally unattainable for intact proteins with masses above 20 kDa. These factors have hindered implementation of protein-centric MS for de novo sequencing of antibodies.…”

“…Recent applications of nMS have highlighted that additional information can be extracted on conformation or folding, topology, and dynamics of various protein complexes, ranging from smaller oligomeric complexes up to intact virus capsids and chaperone complexes 7 . The most-pivotal challenge of nMS is to transfer the higher-order structures and non-covalent complexes to the gas phase without loss of their native quaternary structure 8,9 . There are critical aspects associated with the sample preparation, ionization conditions, and instrumentation to ensure preservation of the structure during ionization 8,9 .…”
