High throughput culture and gametogenesis induction of Laminaria japonica gametophyte clones

Abstract: In anticipation of the application of a new sporelingraising method using gametophyte clones to Laminaria commercial cultivation in China, techniques of mass culture and gametogenesis induction of L. japonica gametophyte clones were developed, as a mass of fertile gametophytes is a prerequisite for sporeling-raising with the new method. Gametophyte clones which were subjected to fed-batch culture exhibited a classical logistic growth curve. Growth rates decreased gradually after 2 months of culture, and were n… Show more

“…Apparently, this so-called free-living technique crucially depends on the propagation of male and female gametophytes under controlled conditions. Zhang et al (2008) reported that temperature (5, 10, and 15°C) had no significant effects on the growth of gametophyte clones which were cultured in 20-L flasks for 15 days and continuously bubbled with filtered air. Li (2003) stated that enhancing temperature could increase the cell number of female and male gametophyte clones.…”

“…Male and female gametophyte fragments were mixed and cultured in suspension in 20-L bottles at 10°C and an irradiance of 60 µmol photons m −2 s −1 in a 10:14 h light-dark cycle (Zhang et al 2008). A sex ratio (female:male) of 3:1 based on the biomass of gametophytes was used for induction of gametogenesis.…”

“…During the expansion and induction of vegetative gametophytes, any operations are strictly disinfected in order to avoid contamination (Zhang et al 2008); no epiphytes were detectable in the collectors during the early culture stage, thus avoiding the need for collectors to be spray-washed. The present results show that most sporophytes grew on the palm ropes via rhizoids once they had grown up to 8-16 rows cells.…”

“…One of the advantageous features of kelps is the capacity of their haploid male and female gametophytes to be multiplied vegetatively through mitosis (Perèz et al 1992;Pang et al 1997;Zhang et al 2008). Though mostly annual in nature, the gametophytes can be grown under strictly controlled conditions for a number of years and be crossed to produce identical sporophytes (Shan and Pang 2009;Xu et al 2009).…”
